Global and Specific Responses of the Histone Acetylome to Systematic Perturbation

Introduction

The histone acetylome is a complex and dynamic epigenetic landscape that plays a critical role in regulating gene expression. Histone acetylation is catalyzed by histone acetyltransferases (HATs) and removed by histone deacetylases (HDACs). The balance between HAT and HDAC activity determines the acetylation status of histones and, consequently, the accessibility of chromatin for transcription factors and other regulatory proteins.

Results

In this study, we used a systematic perturbation approach to investigate the global and specific responses of the histone acetylome to a variety of stimuli. We treated cells with a panel of HAT and HDAC inhibitors and analyzed the changes in histone acetylation using chromatin immunoprecipitation sequencing (ChIP-seq). We found that the global histone acetylome is highly sensitive to perturbation, with both HAT and HDAC inhibitors causing dramatic changes in histone acetylation levels.

Conclusion

Our study provides a comprehensive view of the global and specific responses of the histone acetylome to systematic perturbation. This information will help us to better understand the role of histone acetylation in gene regulation and epigenetic inheritance.
